Hi Hi Puffy AmiYumi follows the adventures of two very cool, but very different pop stars as they travel from gig to gig or just hang out in their tour bus. Being famous rock stars, Ami and Yumi tour the globe in their customized Puffy bus, a veritable condo on wheels with all the major amenities and an ever-changing interior. Ami is the peppy, positive, and resourceful one. Yumi is the hard-rocking, no-nonsense cynic with an absolutely infallible sense of cool. Together, these superstars take the world by storm with musical talent, trend-setting style and humor, despite occasional misjudgments from their well-meaning but tragically square manager, Kaz.

In their childhood years, Ami and Yumi arrive at Camp Youwannasushi and are met by Atchan, who tells them about Rumaki the bunk monster. Yumi is grabbed by Rumaki and Ami and Atchan go under the bed to save her. They manage to save Yumi, but she does not believe them because she was in the sack for the whole episode.
Yumi needs a new guitar and must accomplish certain tasks to get one. So she meets Master Mei Pie, who sends her to get some items. She collects all the items, but realizes that she was not focusing on her playing.
The tour bus is acting erratically and Ami and Yumi go inside to find out what is wrong. They find out that the man who sold them the bus had moved into its carburetor. The girls get him out and the bus is fine.
Yumi is chosen to be a movie star, but Ami has to be her stunt double.
Ami and Yumi have to stop other people seeing an embarrassing photo of Ami in the newspaper.
Ami asks Wall to help out with the band, but he is not good at it.
In prehistoric times, Primitive Ami and Primitive Yumi create their own music, and Primitive Kaz tries to get the two to join together and form a band.
Jeng Keng and Tekirai clone themselves to get rid of Domo after failing the first time.
Yumi is mistaken for a secret agent and must handle the real Agent Y's job.
After coming to life, Ami and Yumi's favorite manga characters battle each other. The fight gets out of hand and it is up to Ami and Yumi to stop them.
While focusing on Puffy AmiYumi, Harmony is disturbed by a member of the Junior Tapeworms and she soon becomes a member and is always getting a badge for every achievement she does.
Ami must fight the Black Knight to see who will defend Kazalot.
When their instruments break, Ami and Yumi go on a quest for golden fleas. They need to find them before the concert is canceled.

The girls go to a prom in the jungle, but are in danger of missing it.

With the help of the girls, Kaz trains to become a sumo wrestler and has his first match.
When Yumi accidentally injures the tooth fairy, she and Ami become tooth fairies for one night. But soon they meet a little boy who would do anything to keep his tooth.
Ami joins Kaz's football team, not knowing that this is a dangerous sport. Kaz tells the security guard not to let Yumi in, because he does not want her to stop Amy playing. Yumi dresses like a cheerleader and gets in. Seeing that Ami doesn't have a chance against the other players, Yumi helps her without Ami realising it.
The show is spoiled because the animators fall asleep. Yumi has Kaz's voice, Ami has Yumi's voice and Kaz has animal voices.
When Ami and Yumi shrink in the drier to mouse size, they need to find a way to get back to normal size while being chased by their own cats.
Kaz tries to find a way to carry Ami and Yumi's instruments to their next gig without the need of a tour bus.
Ami, Yumi, and Kaz get lost in space as they head to their next gig.
When Yumi is struck by lightning, she is sent to an alternate universe where everyone is evil. Meanwhile, Yumi's evil counterpart is put in Yumi's place and wreaks havoc.
Kaz accidentally knocks out a horse named Butterscotch, and Ami and Yumi dress up as the horse to help Kaz out at a rodeo.
An alligator named Waldo appears and startles Ami and Yumi. The girls then try to get rid of him.
Ami and Yumi travel back in time to become disco stars.
The girls are denied access to the Rock Legends museum since they've never trashed a hotel room. Ami and Yumi try their best, but the plan becomes thwarted by an efficient maid.
Ami's cell phone overuse causes Kaz to take her cell phone away, leaving Ami to find alternate methods to communicate with other people.
Kaz gets sent into a manga where he is captured by Mr. Maestro. Ami and Yumi try to rescue their manager with the aid of two heroes, a pencil, and an eraser.
Julie (Lara Jill Miller) comes for a visit. She does nice things to Yumi, but mean things to Ami(Such as calling her Arthur and pretending she doesn't exist). When they go to their concert, Julie is not treated as a star. When the concert is over, she goes to a dark alley and plots revenge.
